    Cultivation of spiroplasma from seasamum affected by phyllody in chick embryos

    A spiroplasma (helical mycoplasma) from the tissues of sesamum plants affected by phyllody disease was successfully cultured in embryonated hen's eggs. The organism adapted to the egg system and grew in the yolk and the allantoic fluid. It readily readapted from eges to as serum enriched cell-free culture medium. Helical, filamentous, pleomorphic bodies were constantly observed in allantoic fluids of inoculated eggs under electron microscope. The egg inoculations triggered faster adaptation of the mollicutes in vitro.

    Distribution and retention of oxytetracycline hydrochloride injected into coconut palms affected by root (wilt) disease

    Oxytetracycline-HCI (OTC) was administered by pressure injector into the trunks of root (wilt) diseased coconut palms. Uptake, translocation and persistence of OTC in various parts of the palms were monitored by microbiological assay of buffer extracts of fresh tissue samples using Bacillus cereus subsp. mycoides (ATCC 6462). Presence of OTC in the foliage was detected within 24 h after administration. The highest concentration was noticed after 5-6 days. Maximum detectable levels in the range of 3-11 Pg/g fresh weight were found in the leaf No. 11. Lower concentrations in the range of 1.0-5 Pg/g fresh weight were detected in the leaf No. 1, 6 and 31 after a week of injection. With the onset of senescence, OTC gets redistributed from older leaves to the most actively transpiring leaves. Very little OTC activity was detected in roots, inflorescence, and spear leaf. Detectable levels of OTC was observed for more than 12 weeks in the foliage.

    Electron microscopic evidence for purification of coconut root (wilt) disease phytoplasma

    Root (wilt) disease phytoplasma was purified from diseased coconut tissues using discontinuous Percoll gradient centrifugation method. Crude sap prepared from coconut was layered on a discontinuous Percoll gradient of 15, 30, 50 and 60%(v/v). After centrifugation at 20.000 gfor 30 min. the turbid fraction formed on the top of 30% gradient in the diseased plant material was recovered, processed and fixed for electron microscopy. Electron microscopic examination of sections prepared from purified preparation of diseased plant material showed typical cells of root (wilt) phytoplasma with heterogeneous sizes and more or less spherical shape that are similar to those found in sieve elements of diseased tissues and salivary glands of infective (viruliforms) . insect vectors. These purified phytoplasma bodies showed trilaminar membrane with internal materials of ribosome granules and DNA fibrils. However, the yield in terms of number of cells was fewer and in addition to intact bodies, free membranes and empty bodies lacking internal contents also were observed.
